Overview
I conduct CRISPR array screens to identify genes whose knock-out promote a specific endpoint of interest. My role consists of growing and splitting cells in culture, inducing Cas9 expression and gene editing through different methods, using high-throughput liquid handling systems to work with 384-wells plate for the screen, as well as electron microscopes for imaging as part of immunofluorescence assays. I also prepared write ups of my experiments for my supervisor.
Skills
I have developed basic skills in wet and dry lab work, such as cell culturing, immunofluorescence, various transfection techniques, gene KOs via a CRISPR-based system, Western Blotting, imaging, data analysis and report writing. I have also been exposed to a variety of projects inside the company from different departments, giving me insight into the nature of roles other than mine.
Responsibilities
After the first few months, I was basically given full autonomy over my work, with the opportunity to regularly ask my supervisor or other colleagues for guidance. However, I was regularly evaluated on my progress through reports and oral presentations to my supervisor and the wider team I belonged to. Moreover, the project I was assigned to was not high-priority.
Support & Guidance
I was in regular contact with both my supervisor and line manager. The atmosphere was very relaxed and I felt comfortable asking questions to anyone within my team. I was given shadowing opportunities as introduction to new techniques, with me attempting them under supervision before continuing on my own as I gained technical confidence.
Culture
Relaxed and open minded, although the size of the team makes it hard to keep in contact with everyone without regularly and proactively reaching out.
